Personality: There are two sides to Ryuuko Matoi.
The first is the most readily apparent, the one you'll meet if she's in a neutral (or worse) mood. Distinctly unladylike, Ryuuko talks like a Yakuza thug and carries herself the same. From the very second she was born, her life has been less than kind to her, and the evidence is apparent in her personality. Originally conceived only to be an expressly unethical experiment to fuse a human's cells with "Life Fibers," then fostered as a weapon to eradicate them, Ryuuko was only recently given the autonomy of her own personhood.
Ryuuko spent most of her life thinking that her mother was dead and spent most of her time at boarding school, getting to know very little of her father that was "too invested in science" to spend time with her. At first, she was bullied for being unable to fit in, until she got fed up and started standing up for herself...with her fists. Unhealthy for sure, but it had results; by middle school she was the head of the punks at her Kanto school...only to turn on them as even they couldn't provide the camaraderie she desperately craved.
It's implied that instead of actually boarding at any of the schools she was sent to, she wandered, living on the streets with only a guitar case to her name, likely the source of her ability to live in the worst conditions (like with the Mankanshoku family in the ghetto) without problem; she refers to herself as a "vagabond", and throughout the series she has moments where she has to keep herself away from others, not necessarily because of a sour mood but because she needs "alone time" in order to destress and collect herself.
The turning point in her life is when her father was murdered. Only then did she realize that she knew nothing about him, and more importantly, that she regretted it. For the first time in her life, she had something to work towards: finding her father's murderer. What starts as a journey to avenge him ends up becoming not only discovery of what he was really spending all that time researching, but the fact that she, along with Senketsu and the Scissor Blade, were all designed from the very things, Life Fibers, that were intending to annihilate the entire planet. She goes from having no reliable identity right into an existential crisis, but comes out of the other end with friends, an adoptive family, and most importantly, a contentedness with who she is.
While many people (read: the Wiki) might describe Ryuuko as stubborn, it doesn't actually fit very well. While Ryuuko is very tenacious about her goals and sees them through, there's almost always something that gets in the way between her and success. Sometimes the adversity is too much, unable to brute-force her way to a solution. When she's backed into a corner, she usually flees in the interest of a better chance of victory later, like many of her early fights in the series. Other times, her emotions get the better of her and she will "run away" from an issue on an emotionally-driven rampage, such as when she went into a berserker rage mid-series, or when she found out the truth about her origins. Her only motivation was to kill and maim, utterly unable to be reasoned with. She does have a lot more control over herself than she used to, but it's not because she's become at all stoic. Even if she might end up falling off the tracks every so often, she manages to get back up and continue fighting for what she wants.
The other half of Ryuuko is the unexpected one. Borderline feminine and cheerful, it's the part of her you really have to dig for if you want to see it. There is a genuine smile and happy laugh that resides in her, but for the most part it stays hidden. She's content to stay guarded, only allowing herself to show this side of herself to people that she either instinctively trusts or has been proven to trust. It takes less to find it at this point in her life than it ever has been before, but that wall of self-preservation still exists.
Compassion for others is a core trait of hers, though it manifests in an unconventional way. For all of her rough, devil-may-care exterior, she does exhibit some common decency--and even go out of her way to show it--for strangers on a one-to-one basis, and definitely for friends and (what she considers) her family. Yet at the same time, she cares very little about "collateral damage"--indirect injuries or losses caused by her actions. Chalk this up either to her drastic emotional swings or her unusual upbringing competing with her natural desire to protect others; it depends on the context of the situation.
In the end, Ryuuko is still a delinquent at heart. Even though the events of canon have helped her discover and come to terms with who (and what) she is, that doesn't change that she spent most of her life as a homeless, vagrant rebel prone to start fights, and the lifestyle she's most familiar with.
